From: SP1 transcriptionally activates HTR2B to aggravate traumatic spinal cord injury by shifting microglial M1/M2 polarization

Fig. 3

HTR2B knockdown promoted the polarization of LPS-induced BV2 microglia from M1 to M2. (A) The effect of HTR2B shRNA in downregulating HTR2B protein expression was determined by western blotting assay. (B-I) BV2 cells were divided into control group, LPS group, LPS + sh-NC group, and LPS + sh-HTR2B group. (B) HTR2B protein expression was determined by western blotting assay. (C) MTT assay was used to analyze cell viability. (D) Tunel assay was performed to analyze cell apoptosis. (E) Western blotting assay was used to detect protein expression of Bax and Bcl-2. (F and G) qRT-PCR assays were conducted to analyze the mRNA expression levels of TNF-α, IL-1β, IL-4 and TGF-β. (H and I) iNOS, CD86, Arg-1 and CD206 protein levels were analyzed by western blotting assay. *P < 0.05
